| 背景描述 | CD28,is a ligand for B7-1 (CD80) and B7-2 (CD86), a well-known co-stimμLatory receptor expressed on T cells. CD28 is a type I transmembrane glycoprotein of the Ig superfamily. When the T cells receive the first signal delivered by APCs (antigen-presenting cells), it initiates signaling cascades, which favor T cell survival and proliferation. It has been demonstrated that patients with lung cancer who responded to PD-1 therapy had more CD28+ T cells, suggesting that CD28 may predict treatment response. |

1. 配置anti-CD3功能性抗体
用PBS稀释配置anti-CD3功能性抗体，浓度为1-5 μg/mL。根据需要选择并计算所需抗体的量。2. 抗体铺板
用稀释后的anti-CD3功能性抗体铺板：
▪ 96孔板：每孔加入50 μL
▪ 6孔板：每孔加入500 μL
37℃ 2h或者4℃过夜（过夜包被效果比较好）。
3. 洗涤
加入细胞前吸去孔内抗体溶液，加入PBS（96孔板200 μL，6孔板2 mL）洗涤2次，去除未结合抗体。4. 准备细胞
取外周血PBMC（步骤省略），也可用分选的T淋巴细胞（也可选择在细胞刺激增殖后进行分选）5. 细胞接种
PBS洗涤细胞1次，用含有以下成分的完全培养基重悬细胞，加入培养板内：
▪ anti-CD28（1-2 μg/mL）
▪ IL-2（10-50 ng/mL）
细胞接种密度：
▪ 96孔板：每孔加入3×105个细胞
▪ 6孔板：每孔加入2-5×106个细胞
6. 细胞培养与观察
培养过程中隔天换液，进行细胞计数并观察细胞状态。
说明：以上CD3和CD28抗体的浓度，可根据自己的需求进行选择，如果培养时间短，希望有很强的活化作用可以适当提高浓度，反之，可降低浓度。虽然高浓度诱导活化能力强，但其诱导凋亡的作用也很明显。
