Y-27632 is a potent, cell-permeable small molecule and selective inhibitor of Rho-associated coiled-coil kinases (ROCK I/II). As an ATP-competitive antagonist, it blocks ROCK-mediated pathways regulating actin cytoskeleton dynamics, cell adhesion, and contraction. Key applications include stem cell culture (maintaining pluripotency), organoid generation (gut, brain, liver, lung) to boost initiation efficiency and passaging survival, and preventing anoikis during cell dissociation or thawing.
